If you mean with cards you can get hold of then not that I know of. If you mean using them yes. There's thread listing the Koikatu games here, a few of the HS games here.
In answer to your School Of Love question no, the game is prerendered. Images and animations are made in Koikatu then put in to the game, it's not done on the fly. Games above are made using Koikatu and HS 2 but not as it's game engine.

The closet thing to that might be Emotions Creators, but I've not tried it myself so could be completely wrong. It's kind of Koikatu but you can make and play simple visual novels and scenes with it.
With visual novels like in the listed ones above they're prerendered (or prerecorded), pictures and animations that can't be customised like the Illusion game they were made using.
