Suddenly my windows 10 cannot run [Ren'Py] type games.. Anyone know how to fix?

It's probably your ant-virus software. Add the game's .exe to your AV whitelist. You may have to reinstall the game afterwards if your AV deleted a file.

I would say AV but also make sure python is installed as well.

I had that issue for a brief while after updating my video drivers. The driver install glitched, with no errors, and I had to rerun it.
If the above AV ideas don't work, update/reinstall your video card driver and reinstall DirectX as well, just to be sure. Either of those can cause the issue you are describing if it is not your AV blocking the programs.
