Hey, how do you do the college bathroom glory hole thingy further than the initial "This is a penis!" touch?

You have to repeat it. I just did it at the same time after class every day by clicking the hole, then using the toilet. Eventually she'll go further. It may also be timed with the story, meaning that she won't do anything until she's progressed to a point in the story where it'd make sense.

I see. I'm pretty sure I had that same first event twice. Getting it again was the reason I stopped going there. Thought there was nothing to see there no more, and any furhter glory hole related activities, if any, would happen via story quest. I'll persist then.
Do you know if it makes a difference when after class? After morning or after afternoon class?

If I recall correctly, there are currently 3 levels to the college bathroom glory hole (but I may be misremembering because it's been a long time):

I don't believe it matters the time. I just tried to stay consistent. Every weekday I'd go to class and then the bathroom. The bathroom doesn't take up a time slot so you don't lose any daytime by using it. I'd suggest doing that as you progress through the story. Eventually it'll happen.

It's like 2 or 3 days after the highest Patron tier gets it. At the same time as the second tier on Patreon.

I didn't like this new update. Please, no hate, but Riley has become corrupted very quickly in this update. She's no longer confused, no longer afraid of being caught.
