For me it's SKA (20-30h), though I hear TLS is even longer.
If we're talking actual written characters and not generated NPCs, Probably TLS again. Or one of those big text-based games like TiTs, CoC or LT. Of course, most of those girls in those text-based games are going to be lesbian futa harpies or some other random shit like that. Some other games that go for very large harems:

RPMG titles like lonargp, caliross, claires quest, SKA, dilmur, violated heroine can be long when you do gameplay and exploration instead of just porn scenes/dialogue.
Theres neverwinter nights mods that are in similar boat with gameplay systems, like a dance with rogues.
Some jap games like Sengoku rance have lots of content because the game part is extensively built up.

My New Family is incredibly long, parts 1-9, then chapter 10 through whatever.
If you sit there and read every bit of dialogue and don't skip through it, then
you're gonna be playing for a while. But you're not doing much content wise
because it's a visual novel only.
