Example:
2 different routes throughout the game with different LI's and slightly different settings but the same MC.
The routes overlap for story relevant scenes but choosing one closes the romantic scenes for the other and vice verse.
Only after finishing booth routes would it be possible to play the third route where the player could be able to choose characters from booth camps within a third slightly different setting.

My personal opinion: as a player I strongly object games/AVNs where you have to replay in order to bed all the LIs. I believe that choices should reflect different "approaches" or "attitudes" towards progressing with LIs, but never force the players to choose between different LIs. And god forbid, never ever should a "bad" choice put the player in a dead end and force them to reload an old save. This is just bad design.

As for different ending, if it's only one scene than this is something I can live with, but for me "save now and replay different path later" turns a fun activity into an accounting chore.

Replace "storywise" by "relevant to my kink/interest", and you get my answer.

A game like Karlsson's Gambit more than one route that interest me, so I follow them in parallel. And a game Heavy Five have a really interesting story, so I don't care too much about the lack of harem possibility. But if the story isn't strongly interesting, or the kink really different and appealing for me, I'll just not care.
Anyway, I'll not replay the game just for this. So, as Doorknob22 said, it will also depend on the game mechanisms. If they don't permit to easily follow more than one playthrough in parallel, I'll just discard the game.

I don't think secret content/routes/endings should be tied to completing all the other endings. It kind of takes away the whole idea of it being 'secret' if you just have to play the game twice.

While some games will point you towards a secret ending after a normal one, it's very different than just being given the secret.

I think College Daze is a great example of how multiple choices/routes should work. There is a ton of content, no matter the choices you make. You can focus on the story or not, you can focus on a few girls or all of them, etc... You'll get a lot of new content every time you replay the game (assuming you don't make the exact same choices).
I really suggest game devs who want to make a game with multiple choices to play this game a few times (even if doing something like this can be very ambitious because it must be a ton of work).
