In my experience: Prime adjusted the stat gains during their intros to make it easier to do stuff with them sooner. I never got first meet beej from Jubes, and I never thought to try to get her to show her boobs.
There is an indirect-quote in this thread that says that Oni will never add Mistique because it would be a pain in the ass trying to get her body morphing mechanics to work. In case you don't know, she is in the Expanded Mod version, but her morphing worked with less than half of the current lineup, and she doesn't [seem to] morph in the most current version of the mod. And the other three are also not happening. Our last three options are

I'm playing the game on Linux (Linux Garuda to be exact).
Linux has a game manager called Lutris. I've woven practically all of my games into it. Also roguelike.
You can run the game under WINE (which is something like a Windows environment simulation under Linux) but that's not necessary with Rogue-Like. The game has a start for Linux that only works for me under Lutris.
Put the game somewhere on the disk and then set it as a native Linux game under Lutris and include the following file as the start file:
/Rogue-Like/Rogue-Like Prime Mod.sh
or only
/Rogue-Like/Rogue-Like.sh
if you play the normal version.
The main thing is the file in the games folder with the .sh at the end.
I might've gotten the mods confused. Maybe I'm thinking of the one from Dittymyman

So, just to check- you can change your gender in this game, but you can't actually change your sex? You need a new save for that?
No, you can just go to Beast's lab and change it there. Also, I found how I can get extra grabby with the Ditty Mod.

I can change my gender in Beast's lab, but I still have jilling off as an option and don't seem to have a dick.

I got them no problem when female zero first released. I'll try again to see if anything's changed.

No need to check. I brought this on myself:
Non-addictive Mode isn't much of a time vampire as male Zero, so apparently more head pats are the answer as female Zero.

I play the non-addicted setting with the prime mod as female zero.
Befriending Laura unlocks the Pain Slut ability. Then you can already give light slaps on the bottom while kissing. After a while, the ladies become pain sluts themselves and are very easy to bring to orgasm. That helps immensely in befriending them.
Gifts from the mail also help immensely. clothes, sex toys and books. Going on dates (just as friends *wink*) also helps push the bars up.
Only in the case of Jean do I advise not to use the love bar, at least at the beginning. Better to base all interactions on loyalty with her. Even if the love value ends up in the negative, it always goes back to zero after a short while.
