Ok, I don't currently have this title installed, so this is from memory and cba to look up the names atm:
Kate
Ophelia (Father quest)
Queen / Princess (requires Front map as well more than likely, but King is ultimately based in City)
Church (see what Priest / Paladin / Syfia say, not just Kirlic Monastery: from memory Paladins are not based in Kirlic)
Cult (Mini-boss Prophet links to City)
Emily 2nd daughter (Cards player / fashion design threads)
Baron's Son's Bride (usually remains in camp or chastity belt, Father is in City)
Dwarves / Dark Elves (very tenuous but requires Mage specialist to even approach area, see their dialogues - so where else are you going to put getting some specific magic training etc?)
Bandits / Revan (see dialogues - if you're ultimately going to replace the Baron, getting Revan pardoned / jail broken will require politics higher than Baron, which means... the City)
Rebel plotline (requires Front map 1st, but ultimately...)
Etc.
For Act II any sane designer would immediately know that something like this is going to be required and plan accordingly (map of Waterdeep)

I mean the building in the SE corner next to the barn. As far as I'm aware this has been present since 4.5ish - the point remains that an early quest to mill flour or clear of rats is somewhat of a tradition in these types of RPGs... but the building has remained stubbornly unused for at least a whole number of patch cycles. This doesn't mean it needs using ASAP, it merely shows that the Dev knows how these hooks work.
Do you have a non-Patreon locked link to this? I'd be genuinely interested in reading over it to see the feasibility.
