So far I've seen nothing new, but I'm speedrunning the areas. If it's a Daycare update, we simply will not have access to that from the posters.
Go through Jesse's Playhouse, and after the sewer portion and past the keycard room go and play the arcade machine inside.

The new content is: Free the people at the playhouse as you normally would (a time consuming and annoying process) and then access the locked door with the keycard to fight DD as a boss.
There's a lot of unique scenes in this fight. Getting grabbed by your counterpart hero, getting grabbed by the boss herself, getting put in the null suit, and getting grabbed by the big machine that drives back and forth. Each of these scenes has a unique variant for whispy or sam.
The game over is the same for both heroes. And the fight is not that entertaining from a gameplay perspective, largely a product of the new combat system's total jank. Recommend getting money at the ATM in the hub world and getting to power level 8 so that you can actually kill things at all in this game.
As far as I know there's shit all else. Enjoy.
