Jess's relationship quest is being added in the next update.
This is usually caused by insufficient permissions. Try running as administrator or moving it to another directory. Your antivirus or anti malware could also be being overprotective.

Great stuff! Is that mandatory or might there be a way to Törn it on or off if someone still wants the ambiguity of choices?

There won't be any way to turn it off, I'm afraid, it'd be too much work to implement.
I absolutely love Missy, thank you, Trinian! Hadn't played in a good whiile and started all over again and loved nearly every second (the start of the Reign of Jess felt a bit extreme and the resolution was too neat, everything else was amazing).
Small bug report, probably it's been mentioned already: I stuck with Poppy all the way against the good girls and ended up stuck, couldn't go to sleep. On the same day that quest finished (with Angel becoming Poppy's new slave) Poppy had invited me to her house to reward me. But that scene wouldn't play because the other scene was still ongoing. Avoided it by loading an older save and delaying the end of the quest by a day.

DIdn't work for me :/ Made all files an exception in my antivirus, and ran as administartor, moved all files too.

You could always try re-downloading the game, but it does sound like overzealous security rather than anything with the game itself.

For my part, I'm looking forward to the chance for Missy and Jess to be bad together...
On the subject of being bad, when it comes to ribbons - which I appear to be mildly obsessed with - I appreciate what you said in answer to a previous question about them being added to older renders gradually, which is fair enough. Will there be ribbon versions of new free roam content at time of release (thinking for example of the screenshots of Missy and Jess in the public post above)?

After having had some sexy times with the slave girl in Missys room, I slept, and wanted to make a save, and the game crashed..
I've found an instance of that quest hanging in the kitchen after the scene in which Missy is forced to wear the slave outfit and do Madeline's work. Is that what you mean?
Didnt even get that far. Its after the Madeline introduction, where Madeline cooks dinner for the family and after that, comes to see Missy in her room, while Missy does her homework. Then you can choose whether to have some fun with her, or send her away. After that I slept, and when I wanted to save my game the next morning, the game crashed..
