The sister can only be drugged when she's on her own during the Weekend, and it also depends on the time of day.You can generally only dose the Mother and sister once a week, since the cooldown is longer than the weekend.
The mother can be drug the morning when she prepare breakfast (sis is not here only during wkend) and sis can be drug when she watch a film (in the third part of the day if i remember correctly)

Hmm, for me the game always had them at breakfast everyday, so I was only ever able to dose them during the weekend.

I love the mind control/corruption aspect, Transformation not such a huge fan of. But the game is good and it actually got a pretty dark story outside of all that. But, each to their own, right.

I played through 5.0.1 Alpha and here is my basic review of the game.
Overall, excellent use of html code to bypass having to use a third-party game engine. Very creative use of html code, genius actually.
Clever and unique story which is engaging and interesting.
My biggest complaint with the game is the choice of models. I fully understand that this is highly objective. I don't find the majority of the models to be attractive. I like Dakota and Tasha (mom is OK) but the rest I don't like at all. I'm not a fan of thick women with fake breasts (and many people aren't, though I can't speak for everyone obviously). Fake breasts are a huge turnoff to me. I'd much rather see a woman with real A cups than fake D cups. Overall, I feel the model selection could have been much better!
The image and video quality are mediocre at best. Many of the images are actually blurry (poor quality image blown up). Many of the videos look like they were filmed using video technology from the 1960's (some are really grainy).
It would be nice to add additional ways/options to dose the various characters in the game. Currently it seems quite limited.
It would be nice to add a phone option to all characters in the game once you achieve a relationship score of 70 or higher. This would allow dates and/or contact to occur with other characters aside from mom, Dakota, and Penny. More hookup options are needed.
Currently, the dates with mom and Dakota are the same each time. It would be awesome to have three to five date scenarios with each character. Going on the same date every time is boring.
This game has tons of potential! I wish the model selection was better and used high quality images and videos. More options to dose and contact the characters in the game would help a ton also.

Well Penny has also real breasts, but you are right there are too many women in this game with the same body type (Julia, Mom, Diana and Sophia). But well its the Artist Choice and we can ignore the women in this game, who not match our tastes.

Yeah, the file shows 606.7 MB but every time I download it, it tells me the download is complete but is only 474 MB.
However, I found a solution to it. I was downloading the file using my browser, before. But then I tried to download the file using the desktop application from MEGA. This time, the file downloaded completely.
