In the Overworld minimap, on the upper right hand corner, you have your three allies under Annabel's photo, click on John so you can take control of him and enter work to speak with Mr. Bastard.

Heya. Id appreciate some help, cuz im stuck before the biker boss. I need to defeat bikers 8 times, but their missions are not appearing at all... Did i miss something?

It's RNG you have to keep doing Night Raids and the other missions.
I'm not sure, but successfully completing the story missions might raise your chances.
