Got it. I completed the quest that unlocks the lab, but ever since then, every quest I’ve started either needs some corruption or is stuck in a loop with a requirement I haven’t figured out yet. I’ve explored every part of the monastery I can access, but now I don’t really see a way to move forward.

Generally quests should either demand higher corruption or be "on hold". If that is the case, the story should progress pretty much on its own. Remember to go into the lab also during the early parts of the game corruption often rises through events with the monks (spending the evenings drinking with them), so maybe you can talk to them through the door or you may have to wait until their event fires.

I get the impression from reviews and comments that the game's main storyline can be played to finish already is that true?

Not completely yet, but it is definitely getting close to the end in the current update as long as their isn't some big reveal or whatever that puts the whole story on new rails.
