Based on when I get it and what the image is, I would say you have to kill 5 of one particular enemy.

I've not opened the update yet... There's no "save" folder so how does it follow on from last version? DO I need to start from the beginning? Or can I copy paste the new "archive" file?

Try playing it. Copies of saves are kept in another folder that stays there even if you delete the game.
