I'm also having trouble getting the brothel mechanic working... After the first one, every time I return the option to work is greyed out. Not sure how to progress forward here.

For the brothel you must reach Ero Value 100. You can either get hit by enemies and raise it or buy a "horny pill" on the pharmacy. Once you reach max Ero value, you should be able to "work" at the brothel.
