I gets unlocked during the Supergirl sequence. The starter on a new day is:
if seensupergirl and seensuperpowergirl and supergirltraining == False
You have seen Supergirl arriving, you have seen the Supergirl/Powergirl scene and have not done training with Supergirl yet.

the variable is locivy
it should be True on the console, check it (shift + o), just type it in. If it's False, force it to be True, but that's really weird if it isn't and you've already met Poison Ivy during Supergirl training.

What specific event are you referring to? Read the message that appears after you click, it might tell you why you lost anyway.
And yes, first encounter with Poison Ivy on that Boobie Trap quest is a loss always. You need to go back again.
