Go to the theatre and ask her for an advice. But you need to complete her event first in order to trigger the option.

Is there a hidden scene in the virgin-only room at Sereme or does it not really matter? I was going to do a virgin-only run but then I went back and ran into an elite thief without really paying attention. Is there that much of a difference in the story and scenes for virgin versus non-virgin run throughout the game besides endings and lewd content?

There's not much of a difference if Reiri's virgin or not, mostly just a bit of a dialogue and that. There's only one ending and Reiri's interactions relies on the lewdness stat specially if she's whoring herself
