If the course button is greyed out you are either arriving too late/too early (the text box SHOULD give you a time frame when to attend) or you have done it already. It's a one time thing, just in case you are trying to do it again

I feel like the Gladiatrix School is completely busted. I've had Akari enrolled there for long enough to definitely trigger one of the 10-20% passive boosts, but the only effect at the end of the day is a loss of affection and health. They're just beating the piss out of her over there :|

What's her happiness? In game text strongly suggests that matters.

What's her stats? How is she looking like? Do you use her as a follower too?

And by your definition what is "long enough" to "definitely trigger" the boosts? You could always just keep reloading the day end screen until it triggers, if it's that important to you.

Also do note that the chances depend on her suitability score, which should be 50 to gain the advertised percentages.
