That really depends on your definitions.
Assuming
no sharing = only 1 partner.
no prostitution = no brothel
Assuming unmodded:
Neutral
Harem *& Ruin* ending ? No. You need Sena & Lena favor, which requires the ranking up of the brothel, which would be "prostitution"
Assuming Complex Breeding:
Neutral
Harem *& Ruin* ending ? Again, no. Same reasons. There is no job in CB that could increase favor with Sena & Lena as a bypass. (No job that increases favor with Amelia, but she shouldn't be a problem regardless)
If brothel usage is fine as long as partners doesn't go beyond 1...
That would be a lot of micromanagement.
(unless non penetrative is fine, tho the game DOES count it)
EDIT: One correction. Since brainwashing requires full favor, the Ruin ending isn't doable in this challenge either.

I see. Thank you for the detailed response. So, with a modded game can I start in unlimited mode and achieve what I asked?

If you mean the free-play mode...doesn't that kinda defeat the purpose of your challenge of beating the game with those restrictions ?
As for starting in free-play mode...I don't know any mods that do that.
CB for dummies lets you modify the cost of the estate, which would make it more reasonable to afford it and reproduce girls. (base cost is 50k)
But that would mean running CB.
Alternatively you could simply give yourself 50k at the start of the 1st day to buy the estate.

Can someone explain tattoo mechanic of elves to me? I've tried searching "tattoo" in this thread but unless I'm misunderstanding something all of them are about mod this, mod that. I don't have any of those mods, I just want to understand how this mechanic works and how to engrave a tattoo to an elf.
Tatoo on Elves are some sort of a binding. When you get the female elf, she has no binding. Once you put her to breeding, the binding is linked to the race of the first male. She then gets a +30% conception chance with the race of the binding. There is a also potion to remove the tatoo (remove the binding).
As an example, I give Claire (Special character which is also an elf) to a goblin. Once she loose her virginity, a tatoo can be seen on her lower belly and on her description, you can see "Tattooed by : Goblin".
Then the trait "Heat" is linked to the goblin race (not only the one who has taken her verginity).
Hope that helps you understand how this mecanics work.
Basically it's +30% conception rate when breeding with the race mentioned.
Let's say the elf has a conception rate of 30. Tattoo is set to Human.
When breeding with anything other than humans, the elves conception rate is 30. But when breeding with humans, it's 60 instead.
The tattoo is set during any interaction between the elf and a partner while the tattoo is not set.
Edit: Interestingly enough. The tattoo removal potion resets the unique partners counter and who the first partner was...as if restoring the virginity. Also works on non-elves in that regard
You can keep some girls for your own use, no problem.
Getting Sena and Lena's approval will require you to prostitute at least one woman. Getting that approval within standard run of 50 days might require more than one prostitute. It's hard to tell how reputation works. But filling one of every kind of room would certainly be enough. If I were you, I would restrict myself to one glory hole room in the brothel and see if it is possible.
You can have any girl outside of brothel have only one unique partner, although the typical strategy is super-incest where a single woman is the partner of her son, grandson, grand-grandson and so on. But you don't really need to do that, if you're willing to dedicate two breeding pairs (with supporting infrastructure) to any monster sale goal you're getting.
Honestly speaking, I find it rather odd you'd want to impose such a challenge and not have considered the roadblocks mentioned.
The game isn't that hard, sure, but that's coming from someone who had beaten it. Some might fare differently.
Normally ppl do self imposed challenges after beating the game on highest natural difficulty, after which one would be expected to know most of the game's quirks.
Most notably favor and the brothel, which are the main roadblocks for this specific challenge if going for harem ending.

So, experiment shows that 4 women (special slave elf and her 3 daughters, with only the mother getting impregnated and giving birth) are safely enough to get Sena and Lena's approval. Reputation levels were not as difficult as "clients guided", which seem to depend on getting clients what they want (which is difficult when not every position in the brothel can be covered).
I challenge somebody to do it with less brothel women.

First, yes, she was the second unlocked special slave.
Second, because Flora the butcher asks you to fuck her up good when she gives you her.
She doesn't have any special traits that would make her better in the brothel.
Sylvia and her daughters would make good money in the brothel, yes. But we're not doing it to earn money, but to impress Sena and Lena. Also, getting Sylvia would normally need a bunch of money to buy all the slaves, as she appears at 66% affinity to Amilia the slave trader.
Flora's own children are a solid option, because they work fast. But they are usually in the lategame, so you'll have a hard time making them work fast enough.
Lena's children could be good, but that requires already having all her favor.
Karen is a good candidate, but you get her from the brothel, so you'll get her as reinforcements, not as the main worker.
I guess that means it's possible to try to run the brothel with Claire the Elf first, then add Karen the Fox Thief and Sylvia the Stripper as they become available, without letting any of their children work in the brothel.
P.S. I also did that playthrough with PunnettRebalance mod. It synergizes well with this kind of restriction, since it does not benefit from super-incest much.
Yeah. I considered Flora, but if Sylvia is considered costly, so would Flora. Even when not considering the estate cost.
