Is it recommended to play the the whole Sequel series in order or is it fine if I jump into Kludge immediately?

You can play Kludge and ignore the others but these games are rich in lore so you will miss hundreds of small things, not anything related to the main story though...
Even if you start with Blight you will miss hundreds of mentions to the other non-porn games by the same dev but even so I recommend you at least play the main series in order.

There's an H scene that I'm missing in the post-game that requires me to go to Tjuerma #2 but the front door is locked... If i progress the main story will I be able to go back?

Tjuerma 2 is always accessible once you unlock it, you go there by going to the garbage dump you were spat out at the beginning of the game
