I can easily say to you that this game dont make anyone gay. I like this gem very much, although i so straight that if i fuck gay guy he will become straight.
In conclusion, not a single media can turn you gay or whatever - if you afraid that game about asses and stuffing asses can turn you gay then this game was not a cause but the consequence.

To elaborate further I was sorta wondering if there was also an in lore explaination for the almost total lack of 100% females (to my knowledge), it seemed kinda interesting.

Some women are born with a cock & balls, some guys are born with asses that don't quit & hips that don't lie. There's no in-universe lore & no apparent societal divide caused by this. Also there's no apparent in-universe stigma that frames getting stuffed by ladies making you gay if you're a guy, which makes it a utopian fantasy that gets past all the labels and focuses on sex, intimacy & fighting.

Why can't the actual female characters, in hetero games, just be female but females who all have large shlongs? Do they have to have vaginas? I'm just a bit curious to hear what your answer might be, for someone asking you that in (genuine) good faith?

There is not, though from what I can tell the lack of females isn't a thing in the lore anyway, the MC is just has penis-magnetism and thus the vast majority of his interactions are with ladies packing meat.

I don't think the world is lacking females, you can find a few actually, but i think Hiro just doesn't meet that many in his adventures.
