The relationship settings are for the patreon no incest rule.

You can change them, if you want to have incest in your VN.
Landlord = Mom or Father
Tenant = Brother or Sister
Neighboor = Uncle or Aunt (most VN's, but here at the Office its more like Sister)
and so on...

i guess you get it.

I would guess, only when you start from scratch. If you want to change them on a "running" game, you could possibly edit them through the Developer Console, but i have no clue about that kind of stuff. Sorry
