Have you defeated all the goblin groups in the south east cave?
That is the trigger to unlock the next area.
Karma and will quests are only optional.

Yes, you need to defeat all groups in the second cave.
Should be 4 groups in there.
Guessing you already got the weakpoint research from the first cave so that the MC can have a chance to rescue grabbed party members.

The plant area? no you don't have to. It is the last room in each area that matters. (the demon dog and mino areas are slightly different in these regards, but only slightly)
