Okay I've searched the whole DLC2 map several times but still couldn't find Asuka and Tama-chan. Anyone knows where to find them?

Asuka is in base game island at one of Big City's building. After defeating her + her scene, you can re-enter her place for an 8-page manga related to this game.
Tama requires you to reenter where her fight occured with Chikeflare.

The last one is No.71, and yes it's the DLC boss. Are you talking about that?
There was an update that added no. 72 which is three different npc characters that Nyuu drew as practice and added into the game just for the sake of it.
There's no boss by the lake, all that's there is Aya + the lake hints.
I've heard that the three girls wasn't properly added to the EN ver maps, switch to JPver in the menu, then go to the lobby of DLC island and you should find three different DLC girls on the beach.
Go to the secret shop (in case you don't know, from Wano shrine, go top right corner to Old road, straight nord there's the shop), buy the toy and go back to the tea house (road between Big City and Harbor Town), back entrance, second floor and speak with the green haired girl.
