I had this weird thing where After making a second stair, I couldn't build anything on the ground floor, which meant I couldn't build anything on the second floor past the rooms on the ground floor. I also couldn't make a third stair, and when I demolished a room, it got rid of the ability to use what I had built there, but I couldn't build anything else on that space. I managed to fix the stair issue when I restarted things, but I assume unless you demolish everything after a room you want gone, you just can't use it again.
