64/32 bit doesn't matter. Save work fine on my W7 32 old laptop fine.64 bit, win 7. In previous versions, the manual saves were working fine. 0.20 and 0.21 arent working. Does it matter where you put the gaming folder? The game did pick up only the auto saves from the older version. So I guess placement doesnt matter.
Good to see Im not the only one. I hope someone can give us things to try and id what the issue is.
Saves are going to end up on the system drive regardless, so probably don't make a difference where you put the game (but always good rule of thumb NOT to put it on the system drive).

And the game does some screwy shit with encoding it based of your windows install ID. So BTW, if you changed OS between plays you're probably toast.
Hence the nasty hack needed to transfer saves (there IS a transfer utility), which is not on the OP due to it's potential issues (I'm assuming).
Anyway, something to try. Make a backup of everything in the mentioned DIR, put it somewhere else. Then delete everything in that save dir. Now start the game and start a NEW game. That should be your only option actually as you don't have any saves now.
See if saving now works. If so, now copy some of the save files back (not the whole save dir, might be some install setup crap in there). Then see if they load/and or you can still save using those loads.
If you want to risk it, you can always new game and use the save transfer util to not have to redo everything. Just RTFM when you use it. It's also probably your only fix if you've upgraded the OS or anything that changes the OS install ID from the sound of it. And iirc, you can possible convert it back by running the save game transfer, load the imported the save in game, change the install ID back to original while the game is running, then make a new save and thus have a save using your original OS install ID.
Edit: A thought, if cleaning out the save dir and start a new game still doesn't allow you to save. Going through the registry and killing anything with Glamour or Sandlust games might be required.
